Background :
SUMO conjugation to target proteins is mediated by a different, but analogous, pathway to ubiquitinylation. UbcH9 is a SUMO conjugating enzyme that mediates the conjugation of SUMO-1 to a number of proteins including RanGAP1, IκBα and PML. It is known to interact directly with proteins that are modified by SUMOylation and may be important for substrate recognition. It appears that conjugation of SUMO-1 to IκBβ prevents subsequent ubiquitinylation and thus protects it from proteasomal degradation. Thus, while ubiquitinylation targets proteins for rapid degradation, SUMO-1 modification acts antagonistically to generate proteins resistant to degradation.
