Background :
Carboxyl-terminus of Hsc70 interacting protein (CHIP) is an E3 ubiquitin ligase. It has been detected in the heart, pancreas, brain, placenta, liver, lung, kidney, and skeletal muscle. CHIP is a multidomain protein containing an N-terminal TPR domain and a C-terminal U box domain both of which may interact with chaperones such as Hsc70 and Hsp70. CHIP is mainly involved with recognizing and marking misfolded proteins for degradation; however, CHIP is a unique E3 protein in requiring chaperone association with target proteins before marking them for ubiquitination. CHIP is therefore important for cell survival as it prevents the accumulation of non-functional proteins in the cell which could potentially produce cytotoxic effects. Previous studies have also suggested that CHIP may be involved in Toll-like receptor (4 and 6) signaling by activating downstream targets such as Src and PKCζ.
