Background :

Tar DNA-binding protein 43 (TDP-43) is a nuclear protein that binds both DNA and RNA at its RNA-recognition motifs and regulates RNA transcription and splicing. It is highly abundant in the lungs, genital tract, spleen, pancreas, and placenta. Tar DNA-binding protein 43 plays an important role in frontotemporal lobar degeneration and amyotrophic lateral sclerosis. In both conditions, Tar DNA-binding protein 43 is found in the cytoplasm of the hippocampus, neocortex and spinal cord, only in a hyperphosphorylated form that is marked for ubiquitin-mediated degradation.
