Background :
CD45, also known as PTPRC, is a receptor-tyrosine phosphatase required for antigen-induced signaling in T-cells and B-cells. CD 45 belongs to the protein-tyrosine phosphatase family, receptor class 1/6 subfamily.It also contains 2 fibronectin type-III domains and 2 tyrosine-protein phosphatase domains. CD45 has been shown to play an important role in T-cell activation through the antigen receptor. CD45 also is suggested to act as a positive regulator of T-cell coactivation upon binding to DPP4. The first PTPase domain has enzymatic activity, while the second one is suggested to affect the substrate specificity of the first domain. Upon T-cell activation, CD45 recruits and dephosphorylates SKAP1 and FYN. Furthermore, it has been shown to dephosphorylate LYN, and modulate LYN activity. Interestingly, CD45 has been associated with several Src-family tyrosine kinases, which are inhibited by tyrosine phosphorylation in their c-termini, suggesting a mechanism for signaling by CD45. CD45 has been shown to bind with GANAB and PRKCSH. Additionally, it interacts with SKAP1 and DPP4, whereas the interaction is enhanced in a interleukin-12-dependent manner in activated lymphocytes. Mutations in CD45 have been associated with Multiple Sclerosis (MS) and severe combined immunodeficiency (SCID), a genetically and clinically heterogeneous group of rare congenital disorders characterized by impairment of both humoral and cell-mediated immunity, leukopenia, and low or absent antibody levels.
