Background :
Damage-Regulated Autophagy Modulator (DRAM) is a lysosomal protein that acts as an effector molecule that is critical for autophagy and p53-mediated apoptosis. In humans, there are 5 known proteins that show significant homology to DRAM. The closest of these proteins is DRAM2, which displays 67% conservation when compared to DRAM. However, unlike DRAM, DRAM2 is not induced by p53 or p73. Additionally, it does not modulate autophagy in contrast with DRAM. DRAM2 is known to be co-localized with DRAM in lysosomes, and co-expression with DRAM is shown to significantly increase cell death.
