Background :
TRAIL-R2 is also known as soluble Death Receptor 5 (sDR5), a single-pass type I membrane protein belonging to the tumor necrosis factor receptor superfamily and is comprised of a single death domain and three TNFR-Cys repeats. Through association of its death domain to cytotoxic ligand TRAIL and related receptor DR4, TRAIL-R2 is a component of the preapoptotic death-inducing signaling complex, the formation of which induces proteolytic activation of caspase-8 resulting in regulated apoptosis. TRAIL-R2 also upregulates the activation of NF-kappa-B. It is expressed in a wide variety of fetal and adult tissues including heart, liver, pancreas, testis, esophagus, prostate, ovary, stomach, spleen, thymus and peripheral blood lymphocytes. It is not expressed in the brain. Defects in TRAIL-R2 are thought to cause head and neck squamous cell carcinomas (HNSCC).
