Background :
Stem cell antigen-1, or Sca-1, is a murine cell surface antigen which is also called Ly6A/E or Ly6D. Sca-1 is essential for HSC differentiation. Once initial differentiation occurs, Sca-1 presence on the cell surface will continue to further influence differentiation; as is seen in skeletal muscle cells that have formed from HSC and which further differentiate into neural and endothelial cells. As such, the identification and characterization of these differentiation cascades might enhance clinical disease therapy by enhancing understanding of the way that Sca-1 is involved in HSC homing and migration. There is evidence to suggest that bone marrow stem cells are signaled by the injured tissue for release into the circulation where they differentiate and proceed to the problem area. Additionally, evidence suggests that aggressive breast cancer tumors express higher levels of Sca-1; that expression of this stem cell marker is up-regulated in cancer. A problem with the migration mechanisms influenced by Sca-1 might result in a prolonged diseased state. The gene encoding this antigen has been cloned independently as TAP (T-cell activating protein) by Reiser et al. (1988), who referred to the encoded protein as Ly6.2. The antigen is expressed on immature hematopoietic progenitor cells and, together with other markers, defines hematopoietic stem cells. Antibodies directed against Sca-1 are used to fractionate early hematopoietic progenitor cells that have the ability to repopulate bone marrow in vivo and are thus considered to represent very early forms of murine hematopoietic stem cells.
