NACHT, LRR and PYD domains-containing protein 3 (NALP3), also called Angiotensin/vasopressin receptor AII/AVP-like, Caterpiller protein 1.1 (CLR1.1), Cold autoinflammatory syndrome 1 protein (CIAS1), Cryopyrin, and PYRIN-containing APAF1-like protein 1 (PYPAF1) is a member of Nod-like receptors (NLR),and forms a caspase-1 activating molecular complex known as the inflammasome. The inflammasome activates IL-1β, the key component of inflammation and fever. NALP3 plays a role in inflammation and immunity and may be a proximal sensor of cellular stress and danger signals. NALP3 protein may function as an inducer of apoptosis and an activator of NF-κB signaling. NALP3 is expressed in blood leukocytespolymorphonuclear cells, osteoblasts, chondrocytes, non-keratinizing epithelia of oropharynx, esophagus and ectocervix and in the urothelial layer of the bladder. Defects in NALP3 have been associated with familial cold autoinflammatory syndrome (FCAS), Muckle-Wells syndrome (MWS), chronic infantile neurological cutaneous and articular (CINCA) syndrome, and neonatal-onset multisystem inflammatory disease (NOMID).
