Background :

MSL-1, also known as Male-specific lethal 1 homolog, Male-specific lethal 1-like 1 (MSL1-like 1), or Male-specific lethal-1 homolog 1, and encoded by the gene MSL1/MSL1L1, is a component of the histone acetyltransferase complex responsible for the majority of histone H4 acetylation at 'Lys-16' (H4K16ac) which is implicated in the formation of higher-order chromatin structure. Binding of MSL-1 greatly enhances MSL2 E3 ubiquitin ligase activity, promoting monoubiquitination of histone H2B at 'Lys-34' (H2BK34Ub). This modification in turn stimulates histone H3 methylation at 'Lys-4' (H3K4me) and 'Lys-79' (H3K79me) and leads to gene activation, including that of HOXA9 and MEIS1. MSL-1 is also up-regulated by gamma-irradiation and MSL-1 interacts with the DNA damage response protein TP53BP1 and the interaction is required for MSL-1’s DNA repair activity but not for its histone acetyltransferase activity. MSL-1 is localized to the nucleus.
