Background :

Histone H4 is one of the 5 main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N terminal tail H4 is involved with the structure of the nucleosomes of the 'beads on a string' structure. Acetylation of histone H4 occurs at several different lysine positions in the histone tail and is performed by a family of enzymes known as Histone Acetyl Transferases (HATs).

Immunogen :

BSA-conjugated synthetic peptide (SG[me2R]GKGGKG-C) corresponding to amino acids 1-9 of Histone H4 with a C-terminal cysteine added to facilitate conjugation. The immunizing sequence is conserved from Tetrahymena to human.

Specificity :

Recognizes methylated Histone H4 (Arg3), MW 10 kDa. Addition of 0.1% Tween®-20 in western blot buffers results in the detection of additional proteins at 17 kDa (possibly methylated Histone H2a), 34 kDa and 80 kDa. Specificity demonstrated by ELISA using methylated and unmethylated Histone H4 peptides. The antibody does not detect recombinant Histone H3 purified from E. coli.
