Background :
Two very important glutamate receptors in the brain are the metabotropic glutamate receptors, mGLUR2 and mGLUR3. mGLUR2 and mGLUR3 work to inhibit adenylate cyclase activity because they operate through the Gi/Go inhibitory G-proteins. Thus they decrease NMDA receptor activity and mediate excitotoxicity and are often targets to attenuate schizophrenia and hyper-excitability brain activity. mGLUR2 and mGLUR3 are primarily localized on presynaptic membranes. They are members of what is termed Group II glutamate receptors and work to reduce the activity of postsynaptic potentials both excitatory and inhibitory in the cortex. Because of their mediating effects synaptic signals, mGLUR2/3 receptors have emerged as exciting targets for novel therapeutic agents used in treating psychiatric disorders. Moreover recent research shows that glutamate receptors can form natural hybrid receptors with other receptors like those for serotonin, and that these heterocomplex formations could be part of the pathophysiology of such brain disorders as schizophrenia.
