Background :
CREB, cyclic AMP (cAMP) response element-binding protein, is a ubiquitously expressed basic leucine zipper transcription factor that functions in glucose homeostasis and stimulates the expression of numerous genes in response to growth factors, hormones, neurotransmitters, ion fluxes, stress signals, and has been implicated in memory because if its apparent role in synaptic plasticity. CREB functions either by itself or with related family members, which include CREB, CREM, and ATF-1, by forming homo- and heterodimers through their basic leucine zipper (bZIP) domains. The CREB signaling pathway is activated by extracellular ligands that bind to cell surface receptors and relay their message through various intracellular second messengers resulting ultimately in the activation of nuclear CREB through phosphorylation of the transactivation domain at serine 1332,4. The classical model of CREB activation is through PKA (cAMP-dependent protein kinase). In the non-active state, PKA resides in the cytoplasm as an inactive heterotetramer of paired regulatory and catalytic subunits. Stimulation of cAMP causes the releases of the catalytic subunits of PKA that passively diffuse into the nucleus and initiates gene expression by phosphorylating CREB at its transactivation domain on serine 1331,2,3,4. CREB mediates the activation of cAMP-responsive genes by binding as a dimer to a conserved cAMP-responsive element (CRE), 5’-TGACGTCA-3’, located within the promoter region of target genes. Target gene activation is terminated by the serine/threonine phosphatases PP-1-mediated dephospho-rylation of CREB.
