Background :
Programmed Death 1 (PD-1, PDCD1 or CD279) is a Type I membrane protein and a member of the extended CD28/CTLA-4 family of T cell regulators. The protein's structure includes an extracellular IgV domain followed by a transmembrane region and an intracellular tail. PD-1 is expressed on the surface of activated T cells, B cells, and macrophages, suggesting that compared to CTLA-4, PD-1 more broadly negatively regulates immune responses. PD-1 may also induce cell death, in association with other factors. Mutations in PDCD1 gene are associated with susceptibility to systemic lupus erythematosus type 2 (SLEB2), a chronic inflammatory disorder of connective tissue. It affects principally the skin, joints, kidneys and serosal membranes. This disease, which is due to PD-1 disfunction, is thought to represent a failure of the regulatory mechanisms of the autoimmune system.
