Hexon is the major capsid protein. The epitope recognized by TC31-9C12.C9 is likely conformation-specific since this mAb can not bind heat denatured hexon. TC31-9C12.C9 immunoprecipitates the complete set of proteins normally associated with intact Ad5 particles; in contrast, TC31-27F11.C2 (a mAb available in the DSHB collection, isolated from the same fusion that generated TC31-9C12.C9) pulls down only free hexon protein. While TC31-9C12.C9 neutralizes Ad5, TC31-27F11.C2 doesn't. TC31-27F11.C2 is the perfect isotype-matched nonneutralizing control Ab for TC31-9C12.C9.
