Samples suspected to contain concentrations higher than the highest standard have to be diluted with Diluent Buffer. Measured results must be multiplied with the dilution factor to obtain corrected results.

The Caffeine ELISA Kit kit is for the quantitative and/or qualitative screening of Caffeine in human serum or plasma samples.

Caffeine, a bitter, white crystalline xanthine alkaloid, functions as a stimulant drug. It occurs in different amounts in seeds, leaves, and fruit of certain plants, serving as a natural pesticide against specific insects. Humans primarily consume caffeine through infusions made from coffee seeds, tea leaves, and various foods and beverages containing kola nut derivatives. Other natural sources of caffeine include yerba maté, guarana berries, guayusa, and the yaupon holly.

This ELISA kit is designed to detect Caffeine based on the principle of "indirect- competitive" enzyme immunoassay. The microtiter wells are coated with antigen. Caffeine residue in the sample competes with the antigen coated on the plate for the antibody. After the addition of enzyme conjugate, TMB substrate is used to show the color. Absorbance of the sample is negatively related to the caffeine residue in it, after comparing with the Standard Curve, multiplied by the dilution factor,caffeine residue in the sample can be calculated.

1. Microplate reader capable of reading absorbance at 450 nm. 2. Automated plate washer (optional). 3. Pipettes and pipette tips. Multichannel pipettes are recommended for a large numbers of samples. 4. Test tubes for dilution. 5. 37°C incubator.

Human serum, Citrate-, EDTA- or Heparin-Plasma. The usual precautions for venipuncture should be observed. It is important to preserve the chemical integrity of a blood specimen from the moment it is collected until it is assayed. Do not use grossly hemolytic, icteric or grossly lipemic specimens. Samples appearing turbid should be centrifuged before testing to remove any particulate material.

1× Washing Buffer: Take 1 vial of 20× Washing Buffer, diluted to 1000 ml with 950 mL deionized water, mix well for later use.

1. Equilibrate the required reagents at room temperature (22~28°C) for 30 minutes. 2. Prepare all reagents as previously instructed. 3. Remove excess microplate strips from the plate frame, seal them, and store them vacuum-packed. 4. Add 50 μl of the standard, samples, or control per well. Add 50 μl of the Antibody Solution per well. A 200-fold dilution of serum or plasma samples is recommended. Users can also set their own sample dilution times according to the verification results. For high value samples, gradient dilution is recommended, e.g. 1:100, 1:200, 1:400,1:800,1:1600. The concentration of caffeine in the sample is equal to the detected concentration multiplied by the dilution. 5. Cover with the provided plate sealer and incubate for 30 minutes at 37°C . 6. Wash the plate 5 times with the 1× Wash Buffer. 7. Add 100 μL of the prepared HRP-Conjugated Antibody Working Solution to each well. 8. Cover with the provided plate sealer and incubate for 30 minutes at 37°C. 9. Repeat step 6. 10. Add 100 μl of TMB Substrate Solution to each well. Cover with the provided plate sealer and incubate in the dark for 5-10 minutes at room temperature (22~28°C). 11. Add 100 μl of Stop Solution to each well. 12. Within 10 minutes of stopping the reaction, measure the absorbance at 450/630 nm using a microplate reader.

Reagents in this kit containing hazardous materials may cause eye and skin irritations. 7. Chemicals and prepared or used reagents must be treated as hazardous waste according to national biohazard safety guidelines or regulations.
