Application Notes :

By Heat: Boiling the paraffin sections in 10mM citrate buffer, pH6.0, for 20mins is required for the staining of formalin/paraffin sections. Other applications have not been tested. Optimal dilutions should be determined by end users.

Integrin alpha-V/beta-3 (ITGAV:ITGB3) is a receptor for cytotactin, fibronectin, laminin, matrix metalloproteinase-2, osteopontin, osteomodulin, prothrombin, thrombospondin, vitronectin and von Willebrand factor. Integrin alpha-IIb/beta-3 (ITGA2B:ITGB3) is a receptor for fibronectin, fibrinogen, plasminogen, prothrombin, thrombospondin and vitronectin. Integrins alpha-IIb/beta-3 and alpha-V/beta-3 recognize the sequence R-G-D in a wide array of ligands. Integrin alpha- IIb/beta-3 recognizes the sequence H-H-L-G-G-G-A-K-Q-A-G-D-V in fibrinogen gamma chain. Following activation integrin alpha- IIb/beta-3 brings about platelet/platelet interaction through binding of soluble fibrinogen. This step leads to rapid platelet aggregation which physically plugs ruptured endothelial surface. Fibrinogen binding enhances SELP expression in activated platelets (By similarity). In case of HIV-1 infection, the interaction with extracellular viral Tat protein seems to enhance angiogenesis in Kaposi's sarcoma lesions.

Background :

ITGB3 (INTEGRIN, BETA-3), also called GP3A, GPIIIa, CD61, is a protein that in humans is encoded by the ITGB3 gene. GP3A is a cluster of differentiation found on thrombocytes. The ITGB3 complex belongs to the integrin class of cell adhesion molecule receptors that share a common heterodimeric structure with alpha and beta subunits. The GP3A gene is mapped to 17q21.32. And the GP3A gene has 14 exons. The 3-prime exon is larger than 1,700 nucleotides and contains the 3-prime untranslated region. The ITGB3 complex mediates platelet aggregation by acting as a receptor for fibrinogen. Although the ITGB3 is expressed on the cell surface at normal levels and is capable of function following extracellular stimulation, it could not be activated via the "inside-out" signaling pathways.
