The clone OKT‐8 is a 32‐34kDa type I transmembrane glycoprotein and a member of the immunoglobulin
superfamily. The OKT‐8 binds with CD8α forms either a homodimer (2 alpha chains) or a heterodimer with CD8β
and is expressed on the surface of cytotoxic T cells, NK cells, cortical thymocytes, and also on some dendritic cells.
CD8 functions as a co‐receptor for T cell receptor and specifically binds to class I MHC for antigen recognition and
T cell activation.
This antibody is routinely tested by flow cytometric analysis. Flow cytometry and other applications were tested during antibody development or are reported in the literature.
Application Information:
Each lot of this antibody has been quality control tested by flow cytometric analysis of human PBMCs. For flow
cytometric staining, the recommended use of this antibody is ≤ 0.5µg per 1x106 cells in 100µl of staining volume
followed by a secondary florescent conjugated anti‐mouse antibody. It is strongly suggested that the antibody
reactivity be empirically titrated for optimal performance in the application of interest.
Concentration :
0.5-1mg/ml, actual concentration vary by lot. Use suggested dilution ratio to decide dilution procedure.

Background :
The CD8 antigen is a cell surface glycoprotein found on most cytotoxic T lymphocytes that mediates efficient cell-cell interactions within the immune system. It is mapped to 2p11.2. The CD8 antigen, acting as a coreceptor, and the T-cell receptor on the T lymphocyte recognize antigen displayed by an antigen presenting cell (APC) in the context of class I MHC molecules. The functional coreceptor is either a homodimer composed of two alpha chains, or a heterodimer composed of one alpha and one beta chain. Both alpha and beta chains share significant homology to immunoglobulin variable light chains. This gene also encodes the CD8 alpha chain isoforms.
