Gene Full Name :
proteinase 3
Synonyms :
Myeloblastin; AGP7; C-ANCA antigen; Leukocyte proteinase 3; PR-3; PR3; Neutrophil proteinase 4; NP-4; P29; Wegener autoantigen; PRTN3; MBN
Protein Name :
Myeloblastin
Protein Function :
Serine protease that degrades elastin, fibronectin, laminin, vitronectin, and collagen types I, III, and IV (in vitro). By cleaving and activating receptor F2RL1/PAR-2, enhances endothelial cell barrier function and thus vascular integrity during neutrophil transendothelial migration. May play a role in neutrophil transendothelial migration, probably when associated with CD177.
Subcellular Localization :
Secreted. Cell membrane. Peripheral membrane protein. Extracellular side. Cytoplasmic granule. Membrane raft. Peripheral membrane protein. Extracellular side.
Tissue Specificity :
Expressed in polymorphonuclear leukocytes (at protein level). Expressed in neutrophils (at protein level). Expressed in differentiating neutrophils.
Background :
Proteinase 3 also known as PRTN3 is an enzyme that in humans is encoded by the PRTN3 gene. This gene is mapped to 19p13.3. PRTN3 is a serine protease enzyme expressed mainly in neutrophil granulocytes. Its exact role in the function of the neutrophil is unknown, but, in human neutrophils, proteinase 3 contributes to the proteolytic generation of antimicrobial peptides. It is also the target of anti-neutrophil cytoplasmic antibodies (ANCAs) of the c-ANCA (cytoplasmic subtype) class, a type of antibody frequently found in the disease granulomatosis with polyangiitis.
