On the basis of bromophenol blue dye, SDS-PAGE Protein Loading Buffer is a kind of 5X concentrated solution, it contains SDS, DTT, Glycerol, bromophenol blue dye, Tris-HCL Loading buffer. It can be used in SDS－PAGE protein loading of the common protein.

1. Mix sample with SDS-PAGE Protein Loading Buffer 5X at the ratio 4:1 (i.e. add 4g sample to 1ml SDS-PAGE Protein Loading Buffer). Denaturize at 100˚C for 3-5 min. Then load and electrophorese. 2. Stop electrophoresis when bromophenol blue dye arrives at the bottom of gels. Notes 1. If SDS sediments appeared when balance SDS-PAGE Protein Loading Buffer 5X to room temperature, try to melt it with warm water. 2. This buffer contains few DDT or mercaptoethanol, so you may encounter a sharp smell. 3. Please operate with gloves in fume hood.

Used in SDS－PAGE protein loading of the common protein.
