Indtroduction :

Alkaline Phosphatase is one of the important makers of immunology and molecular biology. The insoluble chromogenic substrate mainly contains naphthol phosphate and BCIP. BCIP colors fresh blue working with NBT, and applies to immunohistochemistry and various dot detection.

Protocol :

1. After the peroxidase detection, wash fully with TBS 4 times and each time for 5 minutes. Do not use phosphate buffer. 2. Add one drop of chromogenic reagent A and B respectively into 1ml of distilled water and mix evenly. Add it to the sample and colorate in dark for 10-30 minutes. If no background appearing, keep coloration until satisfied. 3. Wash fully with distilled water to stop reaction. Counterstain with nuclear fast red if necessary. 4. Seal sllide with water soluble sealing reagent (Boster can provide). Observe with microscope.
