With the advantages of easy and effective operation, Boster's Protein Silver Staining Reagents are suitable for the SDS-PAGE or non-denaturing PAGE. It can be also used for the silver staining of 2D gel, and the staining gel will be compatible with the subsequent mass spectrometry detection. The sensitivity of silver staining is 100 times as high as that of CBB(Coomassie brilliant blue) staining. It is unnecessary to use the toxic methanol.It can stain 10 pieces of gel (8x10cm).

A. Fixation: immerse the electrophoresis gel into the fixative, put it on the horizontal shaker and vibrate slowly for at least one hour. B. Washing: rinsing it with the deionized water 3 intervals for 30 min . C. Sensitization: remove the deionized water of the above step, add sensitizer to the gel. Shake on the shaker at room temperature for 30 minutes (60-70rpm). D. Washing: rinsing it with the deionized water 3 intervals for 30 min . E. Silver Staining: remove the deionized water of the above step, add silver staining solution. Shake on the shaker at room temperature for 20 minutes (60-70rpm). F. Washing: rinsing the gel with the deionized water for 3 times, 1 min each. G. Color developing: remove the deionized water of the above step, add color development solution. Shake on the shaker at room temperature for 1-15 min (60-70rpm). Stop shaking until the satisfied protein band appears. (Note:1. The bands of the silver staining often appear within 15 minutes, or color developing solutions should be changed. 2. Stop color developing if the background got faint yellow.) H. Stopping: remove color developing solution and add the stop solution. Shake the gel on the shaker at room temperature for 10 minutes( 60-70rpm). I. Washing: rinsing the gel with the deionized water for 3 times, 1 min each. J. Photograph, protect the protein band, and recor. Note 1. The silver staining solution and color development solution in dark. 2. Please follow the datasheet strictly. 3. Take notice of the time of the color development for its fast staining. 4. Precool the silver staining solution and color development solution before use. 5. To avoid pollution, please ensure all appliances are clean. Touching with hands is not recommended. 6. Highly purified deionized water is recommended as wash buffer. The distilled water is better.
