Background :

Klotho is a transmembranprotein type I containing ß-Glucuronidaseactivity (EC-Nummer 3.2.1.31). This enzyme hydrolyzes Steroid-ß-Glucuronides. Reduced production of the peptide was associated with chronic renal failure (CRF) in humans. The Klotho peptide released from the membrane bound isoform was discussed as an anti-ageing peptide (Arking et al., 2002) and may function by inhibiting the signal transduction between Insulin and IGF1. Further hints for the function as an anti-ageing peptide are given by the fact that transgenic mice overexpressing Klotho live longer than the related wildtypes (Kurosu et al., 2005)
