Human SLPI Activity Assay Human SLPI activity is measured by its ability to inhibit cleavage of a fluorogenic peptide substrate MeO-Suc-AAPV-AMC by human neutrophil elastase. The potency of hSLPI is described as IC 50 value. Materials 1. Recombinant human SLPI (Predicted MW: 13.2 kD) 2. Assay Buffer: pH 8.0, 50 mM Tris, 1.0 M NaCl, 0.05% Brij-35 3. Human Neutrophil Elastase (HNE; Athens Res Tech, Cat # 16-14-051200; 200 ug/mL stock in 20 mM Sodium Citrate and 50% glycerol at pH 4.5) 4. Elastase substrate: MeO-Suc-AAPV-AMC Activity Assay Procedure . 1. Dilute hSLPI in 8000 ng/mL in Assay Buffer. 2. Make serial dilution of hSLPI in Assay Buffer from 8000 ng/mL to 3.13 ng/mL, and include 0 ng/mL as a control. 3. Dilute human elastase in 2 ug/mL in Assay Buffer. 4. Add into a tube 20 uL of each serially-diluted hSLPI, including the control without hSLPI. Then add 20 uL of 2 ug/mL HNE. Briefly vortex, and then centrifuge the tubes to mix the solutions. 5. Incubate the mixture at room temperature for 30 min. 6. Add 160 uL of Assay Buffer into each tube prepared in step 4 after incubation to make diluted assay mixtures. 7. Dilute the substrate in Assay Buffer at 200 uM. 8. Load into a well plate 50 uL of each assay mixture and start the reaction by adding 50 uL of 200 uM Substrate. Include a substrate blank containing 50 uL Assay Buffer and 50 uL of 200 uM Substrate. 9. Read at excitation and emission wavelengths of 380 nm and 460 nm, respectively, in kinetic mode for 5 minutes. 10. Analyze the data with non-linear regression to estimate IC 50 value of hSLPI towards HNE. Final assay conditions per well: . HNE: 10 ng (100 ng/mL) hSLPI: 400, 200, 100, 50, 25, 12.5, 6.25, 3.13, 1.57, 0 ng/mL Substrate: 100 uM . BioLegend carrier-free recombinant proteins provided in liquid format are shipped on blue-ice.
