Human BACE1 Activity assayHuman BACE1 activity is measured by its ability to cleave a fluorogenic peptide substrate Mca-SEVNLDAEFRK(Dnp)RR-NH2. The increase of the product is monitored by increase in intensity of emission at 405 nm with excitation at 320 nm.Materials1. Assay Buffer: pH 4.0, 50 mM Citrate, 0.1% BSA2. Recombinant Human BACE13. BACE Substrate: Mca-SEVNLDAEFRK(Dnp)RR-NH2Activity assay procedures1. Dilute hBACE1 in the assay buffer at 10 µg/mL (10 ng/µL).2. Dilute the substrate at 10 µM in Assay Buffer.3. Load into a well plate 50 µL of the 10 ng/µL hBACE1 and start the reaction by adding 50 µL of 20 µM Substrate. Include a substrate blank containing 50 µL of Assay Buffer and 50 µL of 20 µM Substrate solution.4.
