Bioassay Protocol Human CRP was immobilized on an ELISA plate at a concentration of 2 ug/mL (100 uL well with a coating buffer of 50 mM Tris, 150 mM NaCl, 1 mM CaCl 2 , and pH 7.5). The wells were blocked with coating buffer that contained 3% BSA before being used in the binding assays. The binding assay was performed with various concentrations of hAPCS-biotin using coating buffer containing 3% BSA at 4 C overnight. Wells were washed with coating buffer containing 0.3% BSA four times. Avidin-HRP at 1:2000 in coating buffer containing 0.3% BSA was used to detect hAPCS-biotin bound to immobilized hCRP. .
