Component:20 μL of 500X Lysosome Probe Green (-20°C, protected from light, avoid freeze/thaw cycles)Required Materials Not Included:Buffer: PBS, phenol-free media, or HBSSDetection/Imaging Guidelines:Ex/Em = 501/509 nm Fluorescence microscope filter set: FITCFlow cytometry channel: FITCAssay Protocol:;Thaw the 500X Lysosome Probe and allow it to reach room temperature.;Prepare lysosome staining solution by diluting the Lysosome Probe 1:500 in your buffer of choice. Aliquot to avoid freeze thaw cycles.Note: Probe concentration may need to be adjusted according to cell type and assay conditions.;Optional: Include live-cell nuclear counterstain such as DRAQ5™ (Cat. No. 424101).;Add an equal volume of Lysosome Staining solution to cell culture media.;Incubate the cells at 37°C for 30 minutes.;Wash cells by twice with PBS or HBSS. If cells are in suspension, spin down and resuspend in buffer of choice.;Add desired volume of PBS or HBSS to cells for observation.;Proceed immediately to analysis.;
