Oroxylin A, also called 6-Methoxybaicalein, is an antagonist at the benzodiazepine site of the GABA(A) receptors.Oroxylin A is an active O-methylated flavone compound that was originally isolated from the medicinal plant Scutellaria baicalensis. Oroxylin A was shown to have multiple functions, including anti-inflammatory, anti-cancer and anti-thrombotic activities. In addition, it has dopamine reuptake inhibitor activity and an inhibitory effect on nuclear factor-κB activation. The compound was used to treat pulmonary infection in Korea, China, and Japan1,2.GABA(A) receptors are members of the Cys-loop superfamily of ligand-gated ion channels. They mediate GABA action, a major inhibitory neurotransmitter, through the central nervous system. The GABAA receptor is considered to be a major target of antiseizure drugs3.

A flavonoid antagonist at the benzodiazepine site of the GABAA receptors. Inhibits [3H]Flunitrazepam binding to rat cerebral cortical membranes (IC50 = 1.09 µM), and selectively abolishes the anxiolytic, myorelaxant and motor incoordination, but not the sedative and anticonvulsant effects elicited by diazepam in mice1.

Centrifuge the vial before adding solvent (10,000 x g for 5 minutes) to spin down all the powder to the bottom of the vial. The lyophilized product may be difficult to visualize. Add solvent directly to the centrifuged vial. Tap the vial to aid in dissolving the lyophilized product. Tilt and gently roll the liquid over the walls of the vial. Avoid vigorous vortexing. Light vortexing for up to 3 seconds is acceptable if needed. For long-term storage in solution, we recommend preparing a stock solution by dissolving the product in sterile water at a concentration of at least 0.1 mg/mL. Divide the stock solution into small aliquots and store at -20°C. Before use, thaw the relevant vial(s) and dilute to the desired working concentration in your working buffer. It is recommended to prepare fresh solutions in working buffers just before use. Repeat freeze-thawing may result in loss of activity.

Up to 50 mM in DMSO. Centrifuge all products before reconstitution (10000 x g 5 min).

Up to four weeks at 4°C or three months at -20°C. It is recommended to aliquot reconstituted product for storage.
