Etbicyphat, also called trimethylopropane phosphate, is a synthetic convulsing compound that acts as a potent antagonist of GABA(A) receptors. TMPP acts through binding to the picrotoxinin and benzodiazepine receptor sites of the GABA(A) ionophore complex. TMPP is eliminated through the urine with no evidence of phase I or phase II metabolism and shows rapid clearance from the blood system1,2.Several studies showed that infusion of TMPP into the nucleus accumbens of adult rats induces subclinical seizures and hyperlocomotor activity3.GABA(A) receptors are members of the Cys-loop superfamily of ligand-gated ion channels. They mediate GABA action, a major inhibitory neurotransmitter, through the central nervous system and play an important role in inhibiting cell excitation in the central nervous system. The GABA(A) receptor is a major target of antiseizure drugs4.

Etbicyphat, an ethyl bicyclophosphate organophosphorous compound, is a GABA receptor antagonist (KB = 3.98 µM for GABA(A) receptors),1 a blocker of neuronal voltage and GABA dependent Cl- currents, and a potent convulsant ( ≥0.4 mg/kg IP in rats) 2,3.
Reconstitution and Solubility :
Centrifuge the vial before adding solvent (10,000 x g for 5 minutes) to spin down all the powder to the bottom of the vial. The lyophilized product may be difficult to visualize. Add solvent directly to the centrifuged vial. Tap the vial to aid in dissolving the lyophilized product. Tilt and gently roll the liquid over the walls of the vial. Avoid vigorous vortexing. Light vortexing for up to 3 seconds is acceptable if needed. For long-term storage in solution, we recommend preparing a stock solution by dissolving the product in sterile water at a concentration of at least 0.1 mg/mL. Divide the stock solution into small aliquots and store at -20°C. Before use, thaw the relevant vial(s) and dilute to the desired working concentration in your working buffer. It is recommended to prepare fresh solutions in working buffers just before use. For long-term storage of diluted solutions, we recommend adding 0.1% BSA. Repeat freeze-thawing may result in loss of activity.
