bcl-2 is from a genomic clone isolated from a normal human fibroblast library. To construct P1+P2Neo, a 279-bp polymerase chain reaction (PCR) product including the bcl-2 5'-untranslated region sequence from just 5' to the AccI site through base -1, relative to the initiation codon, (designated AccI/-1), was cloned into pBluescript KS(-) (Stratagene), using primergenerated 5' XhoI and 3' HindIII sites. Next, a 486-bp XhoI-AccI fragment from the 5'-untranslated region was cloned into this plasmid, producing a 756-bp XhoI/-1 (HindIII) clone. Addition of an approximately 2.2-kb XhoI fragment to this construct created a 3.0-kb bcl-2 P1+P2 clone. Finally, a 2.3-kb HindIII-BamHI fragment from RSVNeo (20) including the neomycin phosphotransferase (neo) gene and SV40 early splice and polyadenylation signals was inserted into the P1+P2 clone to produce P1+P2Neo.
