A 3.5kb EcoR1 fragment was used as the 39 homology arm and subcloned into a modified version of pPNT with a Not1 site introduced into the Sal1 site for linearization. A tau-lacZ-PGKneo cassette (generous gift of Peter Mombaerts) was inserted between these two arms to give the vector described in Kloetzli et al. This deletion construct results in the replacement of the entire coding region of Foxb1 with the tau-lacZ fusion gene and the PGKneo cassette. The resulting mutation is a null. |
