growth notes :
sh1368 sequence= GACGAGAACAGTTGAAACA The FUGW modified vector was obtained from Dr. Valera Vasioukhin at FHCRC (Lien et al., 2008). To generate the lentiviral vector for production of shRNA, the FUGW vector (provided by D. Baltimore, California Institute of Technology, Pasadena, CA; Lois et al., 2002) was modified by placing internal ribosome entry site neomycin sequences downstream from EGFP and cloning of human H1 promoter followed by unique BamHI EcoRI sites upstream from the ubiquitin promoter. Please note that the insert was cloned into BamHI-EcoRI sites using BglII-EcoRI fragment. The BamHI site was destroyed after ligation. See the attached document for a map from Lien et al., 2008 for vector information.
