The 3' primer contains the coding sequence for the HA epitope YPYDVPDYA, the stop codon and a HindIII site. HindIII-cleaved PCR products were ligated into pTet-Splice. The plasmid contains a CMV promoter under control of regulatory sequences from the tet operon such that, when cotransfected with the transactivator pTet-tTAk (Life Technologies), it expresses protein only in the absence of doxycycline.
