Background :

NFM-NM-:B p65 (Nuclear factor NF-kappa-B p65 subunit also known as NFM-NM-:B p65 or RelA) is a member of the Rel family of transcription factors that are involved in processes such as inflammation, immunity, differentiation, cell growth, tumorigenesis and apoptosis. The Rel family includes c-Rel, RelA (NFM-NM-:B p65, NFM-NM-:B3) and RelB (I-Rel). Rel proteins form dimers with the NFM-NM-:B p50 (proteolytically processed from p105/NFM-NM-:B1)and p52 (proteolytically processed from p100/NFM-NM-:B2) members of the NFM-NM-:B family that bind DNA and activate transcription. The NFM-NM-:B p65/p50 heterodimer is the most abundant form of NFM-NM-:B.NFM-NM-:B signaling is negatively regulated by the sequestration of the NFM-NM-:B complex in the cytoplasm by its association with the IM-NM-:B family of inhibitory proteins. The IM-NM-:B Kinase (IKK) complex is the key enzyme involved in the activation and translocation of NFM-NM-:B. The IKK complex is composed of two catalytic subunits (IKKM-NM-1 and IKKM-NM-2) and a regulatory subunit (IKKM-NM-3). Upon its activation, the IKK complex phosphorylates IM-NM-:B proteins (IM-NM-:BM-NM-1, IM-NM-:BM-NM-1 phospho Ser32, 36), which marks them for ubiquitination and degradation. This enables the NFM-NM-:B complex to translocate to the nucleus where it regulates transcription by binding to DNA.
