Flow cytometry analysis of phospho-p38 MAPK (pThr180/Tyr182) on Jurkat cells, either left untreated (blue histogram) or treated with anisomycin (green histogram). Cells were harvested, fixed with formaldehyde for 10 minutes at 37ºC, permeabilized with ice-cold methanol at a final concentration of 90% for 30 minutes on ice, and incubated with a phospho-p38 MAPK (pThr180/Tyr182) monoclonal antibody (Product # MA5-15182) or nonspecific rabbit monoclonal antibody control (red histogram) at a 1:25 dilution for 1 hour at room temperature. For flow analysis, a 30-minute incubation with a fluorescently-conjugated anti-rabbit IgG secondary antibody was performed and data were acquired on a flow cytometer.
Western blot analysis of phospho-p38 MAPK (pThr180/Tyr182) was performed by loading 20ug of HEK293T whole cell lysates, either left untreated (UT, left lane) or incubated with alkaline phosphatase (AP, right lane), per well onto an SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat dry milk in TBST for 1 hr at room temperature. The membranes were probed either with a phospho-p38 MAPK (pThr180/Tyr182) monoclonal antibody (Product # MA5-15182) at a dilution of 1:1000 (top blot) or a total p38 MAPK monoclonal antibody (bottom blot) overnight at 4°C, washed in TBST, and probed with a peroxidase-conjugated goat anti-rabbit IgG at a dilution of 1:40,000 for 1 hr at room temp. Immunofluorescent analysis of phospho-p38 MAPK (pThr180/Tyr182, green) in UV light-treated HeLa cells. Cells were fixed in 4% formaldehyde for 15 minutes at room temperature, rinsed with PBS, and blocked with 5% normal goat serum in 1X PBS containing 0.3% Triton X-100 for 1 hour. Cells were probed with a phospho-p38 MAPK (pThr180/Tyr182) monoclonal antibody (Product # MA5-15182) at a dilution of 1:100 overnight at 4ºC, washed with PBS, and incubated with a fluorescently-conjugated anti-rabbit IgG secondary antibody for at least 1 hour at room temperature. F-actin (red) was stained with fluorescently-conjugated Phalloidin. Staining was visualized by confocal microscopy.
