Western blot using Rockland's Affinity Purified anti-Sts-1 antibody shows detection of a band ~70 kDa corresponding to mouse Sts-1. Approximately 1.0 ?g of recombinant (truncated) Sts-1-GST was separated by SDS-PAGE and transferred onto nitrocellulose. After blocking the membrane was probed with the primary antibody diluted to 0.3?ug/ml overnight at 4° C followed by washes and reaction with a 1:10,000 dilution of IRDye800 conjugated Gt-a-Rabbit IgG [H&L] MX (611-132-122). IRDye800 fluorescence image was captured using the Odyssey® Infrared Imaging System developed by LI-COR. IRDye is a trademark of LI-COR, Inc. Other detection systems will yield similar results.

Detection of Human and Rat UBASH3B/STS1 by Western Blot. Western blot shows lysates of HCT-116 human colorectal carcinoma cell line, HEK293 human embryonic kidney cell line, MOLT-4 human acute lymphoblastic leukemia cell line, U-118-MG human glioblastoma/astrocytoma cell line, U2OS human osteosarcoma cell line, and Y3-Ag rat myeloid cell line. PVDF Membrane was probed with 2 ╡g/mL of Human/Rat UBASH3B/STS1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF6696) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016 ). A specific band was detected for UBASH3B/STS1 at approximately 72 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1 .
