Can you tell me if the amino acid sequence it targets is PSIRCSSVS( aa417-425 in Human SMAD3 and Zebrafish smad3a)? Also, will it recognize/target zebrafish smad3b or smad2? Do you have any independant publications in which this antibody is used in any species, but preferable zebrafish?

Thanks very much for your question regarding antibody LS-C18974 (SMAD3 Rabbit anti-Human Polyclonal (aa417-425) Antibody). Yes, the customer is correct. This antibody is raised against a synthetic peptide corresponding to amino 417-425 of human SMAD3 protein according to P84022. The sequence is indeed PSIRCSSVS and is 100% identical/homologous with the corresponding zebrafish SMAD3a sequence according to Q8AY15. A BLAST analysis of the peptide against the zebrafish SMAD3b sequence according to Q8AY16 shows 78% identity/homology with 7 out of 9 identical residues. A BLAST analysis against zebrafish SMAD2 according to Q9I9P9 shwos 78% identity and 89% homology.
Based on these numbers we would expect to see at least some cross-reactivity. We would caution, however, that in our experinece cross-reactivity in non-target species varies, especially in assays such as IHC that require a bit more affinity from the product. In the absence of testing data we cannot guarantee that the antibody will detect the zebrafish protein. For this reason we list zebrafish as a predicted reactive species.
