Background :
BACE (beta-secretase, beta-site APP-cleaving enzyme) is a 70 kDa membrane associated aspartyl protease that is involved in the two step cleavage of amyloid beta-peptide in the development of Alzheimer's disease. Once synthesized as a proteolytically inactive precursor form, the BACE protein undergoes post-translational modification in the ER, losing the pro-peptide and becomes glycosylated. The modified, mature protein is transported to the Golgi and then continues its migration to the membrane. Once in the membrane, BACE functions as the major source of beta-secretase activity. BACE2, a homolog of BACE, has also been identified. BACE2 differs from BACE in that it is highly expressed in multiple tissues including colon, pancreas, and prostate rather than being restricted to the brain.
